Abstract—— The effects of short-term oral administrationof red wine polyphenolic compounds on hemodynamic parametersand on vascular reactivity were investigated in rats. Endothelialfunction and vascular smooth muscle contractility were studiedin association with the induction of gene expression in thevascular wall. Rats were treated daily for 7 days by intragastricadministration of either 5% glucose or red wine polyphenoliccompounds (20 mg/kg). Administration of these compounds produceda progressive decrease in systolic blood pressure, which becamesignificantly different on day 4. Aortas from rats treated withred wine polyphenolic compounds displayed increased endothelium-dependentrelaxation to acetylcholine that was related to increased endothelialNO activity and involved a mechanism sensitive to superoxideanion scavengers. However, no increase in whole-body oxidativestress has been observed in rats treated with red wine polyphenoliccompounds, as shown by plasma glutathione assay. Also, in theaorta, red wine polyphenolic compounds increased the expressionof cyclooxygenase-2 and increased the release of endothelialthromboxane A2, which compensated for the extraendothelial NO–inducedhyporeactivity in response to norepinephrine, resulting fromenhanced inducible NO synthase expression. The present studyprovides evidence that short-term oral administration of redwine polyphenolic compounds produces a decrease in blood pressurein normotensive rats. This hemodynamic effect was associatedwith an enhanced endothelium-dependent relaxation and an inductionof gene expression (of inducible NO synthase and cyclooxygenase-2)within the arterial wall, which together maintain unchangedagonist-induced contractility. These effects of red wine polyphenoliccompounds may be a potential mechanism for preventing cardiovasculardiseases.
