Gene therapy may help Hemophilia A
WASHINGTON, July 3 (UPI) -- Hemophilia A (Factor VIII deficiency) may be curable with a new gene therapy that is successful in mice.
Writing in the July issue of the Journal of Clinical Investigation, the authors explained that Factors VIII and IX are blood clotting proteins. One in 5,000 males lacks Factor VIII and develops hemophilia A, and one in 25,000 lacks Factor IX and develops hemophilia B or Christmas disease. Both sets of patients will bleed uncontrollably when injured.
Patients are treated with intravenous synthetic Factor VIII or IX two to three times a week; but around a third of hemophilia A patients develop antibodies to synthetic FVIII that inactivate its clotting properties. Treatment with FVIII products that don't elicit this response can cost up to $1 million per year.
The researchers, led by Qizhen Shi and Robert R. Montgomery of the Medical College of Wisconsin and the BloodCenter of Wisconsin in Milwaukee, took bone marrow stem cells from mice with hemophilia, inserted synthetic FVIII into them, then helped the cells develop into platelets.
The new platelets were accepted easily into the bloodstream, where the FVIII remained hidden until the platelet stuck to a damaged blood vessel and released its healing proteins. Mice given 5,000 to 10,000 times the amount of antibody necessary to neutralize FVIII in humans still clotted normally if they had received the engineered platelets. Human trials of the technique will begin if tests in larger animals are successful.
