Protein cutback stops Alzheimer's progress in mice

CHICAGO - Reducing levels of a protein called tau helped arrest Alzheimer's disease symptoms in mice, a finding that might lead to new ways of fighting the incurable brain disorder, researchers said on Thursday.

"If this strategy also works in humans, it could enable a major leap forward in our ability to treat and prevent devastating neurological diseases," Dr. Lennart Mucke of the University of California, San Francisco, said in a statement.

The researchers, whose work appears in the journal Science, studied mice genetically modified to mimic the symptoms of Alzheimer's disease.

They found that reducing production of tau prevented seizures and memory deficits related to the disease, which is marked by a steady loss of memory that robs people of their ability to take care of themselves.

"Amazingly, even partial reduction of tau prevented memory problems and premature deaths in our Alzheimer mice," said Dr. Erik Roberson, assistant professor of neurology at UCSF.

Although Alzheimer's affects the brains of mice and humans differently, the finding is striking, the researchers said.

Most current efforts to treat Alzheimer's disease are focused on ways to reduce levels of amyloid-beta proteins, which build up to abnormal levels and are widely suspected of causing the disease.

In a normal brain, tau regulates the internal stability of brain cells. In patients with Alzheimer's, the protein forms into clumps or tangles. Many treatment approaches aim to get rid of these tangles, but Mucke's study suggests regulating levels of tau instead.

"We wanted to pursue a complementary strategy and try to make the brain more resistant to amyloid-beta proteins," Roberson said.

His team found that reducing tau protects the brain against overstimulation, a finding that might have implications for other neurological diseases.

Alzheimer's affects more than 5 million people in the United States, where it is the seventh-leading cause of death. Current drugs can slow the progression of Alzheimer's, but they eventually stop working.
